“So far, the only adoptive cancer immunotherapy that has been proven clinically to work for treating patients with solid cancer tumors is TIL (tumor infiltrating lymphocyte) therapies albeit with limited efficacy with the exception of melanoma and cervical cancer. “TILs and Fucosylation as separate products has been proven safe and efficacious in Phase 2 clinical trials. Our FucoTIL product is essentially a combination therapy of these two clinically proven products where we are leveraging the synergistic power of TILs with their antigen diversity required for solid cancer tumor killing and our fucosylation TIL efficacy multiplier product which superboosts TIL solid cancer tumor killing effectiveness with the goal of eradicating patients’ cancers” said Lynnet Koh, CEO and Founder of TargaZyme.

For patients with solid cancer tumors undergoing adoptive cell therapies, higher numbers of adoptive immune cells with antigen diversity reaching the patients’ tumor sites are correlated with improved clinical outcomes. Efficacy outcomes for the current generation of TIL therapies are constrained by the inefficient trafficking or homing of the immune cells from IV infusion through the vasculature and eventual entry into tumor sites where today, less than 3 percent of infused immune cells reach tumor sites with minimal intra-tumoral penetration. TargaZyme’s cell efficacy multiplier product enables 250-500% more TILs to be ‘delivered’ from the vasculature to the tumors with a correspondingly multi-fold increase in intra-tumoral penetration essentially enabling the increased numbers of TILs, i.e. ‘soldiers’ to mount more effective attacks against the solid cancer tumors for their obliteration. Interestingly, TargaZyme cell efficacy multiplier technology also supercharges FucoTILs’ solid tumor-killing machinery by enabling the increased expression of Granzyme B, Perforin, and FASL, thus improving the immune response as the cell synapse with solid tumors, all of which contributes to the increased potency of Targazyme’s FucoTILs to more effectively eradicate solid cancer tumors.

TargaZyme’s most clinically advanced Phase 3 Special Protocol Assessment TZ 101 product also targets late-stage patients although with different diseases such as blood cancers, serious autoimmune diseases, blood disorders. An off-the-shelf product that is simple to use and administer at ‘bedside’, TZ 101 has demonstrated its potential in Phase 2 clinical trials to potentially disrupt the $50 billion per year cancer supportive care industry by potentially preventing/reducing the incidence of multiple life-threatening side effects associated with intense chemotherapy/hematopoietic stem cell transplantation.

Today, a patient receiving a hematopoietic stem cell transplant aimed at curing late stage blood cancer, serious autoimmune diseases such as multiple sclerosis and lupus, blood disorders such as sickle cell anemia and thalassemia will receive a vast (over ten, easily 20+) cocktail of drugs (Neulasta, Nplate, anti-viral drugs, antibiotics, anti-fungal drugs, anti-acute GvHD drugs, anti-cGVH drugs, medicine to treat low red blood cell count, medicine to treat internal bleeding, drugs to treat veno-occlusive disease, drugs to treat engraftment failure, various organ failure, etc. all with their associated costs and side effects, just to treat the various side effects of standard-of-care treatment. Phase 2 clinical trials and preclinical testing data at MD Anderson Cancer Center have demonstrated the potential of TZ 101, i.e. just one drug candidate to accelerate both myeloid and lymphoid cell recovery post intense chemotherapy and reduce the risks of various life-threatening side effects associated with hematopoietic stem cell transplantation, such as viral, bacterial and fungal infections, GvHD versus host disease, hemorrhaging, engraftment failure rate, etc.

TZ 101’s competitive advantages include its demonstrated potential to help prevent multiple post-transplant life-threatening morbidities versus competitive products that deliver efficacy for a single morbidity, with its remarkable simplicity with ease of use and administration. This innovative solution enables treating diverse cell types within a mere 30-minute timeframe, at room temperature.
